广西斑地锦及其混淆品DNA条形码分子鉴定

Identification of Euphorbia maculata L. and its adulterants in Guangxi based on DNA barcode technology

摘要: 为鉴定斑地锦(Euphorbia maculata L.)及其混淆品,以广西采集的41份斑地锦及其混淆品为研究对象,提取其基因组DNA,对各个样品的条形码rbcL和psbA-trnH序列进行扩增并测序,与GenBank下载的13份斑地锦及其混淆品rbcL和psbA-trnH序列进行了分析比对,计算了斑地锦及其混淆品的种内、种间遗传距离并采用邻接法构建了系统发育树。结果发现,斑地锦种内遗传距离明显小于其与混淆品的遗传距离,系统发育树中斑地锦样品聚为单系并能够与其混淆品明显区分开,这表明rbcL和psbA-trnH序列可用于斑地锦及其常见的混淆品的鉴别。

Abstract: In order to identify the species of Euphorbia maculata L. and its adulterants,the total genomic DNAs were extracted from 41 samples of Euphorbia maculata L. and its adulterants from Guangxi, and the barcode rbcL and psbA-trnH sequences of each sample were amplified and sequenced, and compared with the rbcL and psbA-trnH sequences of 13 samples downloaded by GenBank. The intraspecific and interspecific genetic distances of Euphorbia maculata L. and its adulterants were calculated and the phylogenetic tree was constructed by the neighbor-joining method. The results showed that the intraspecific genetic distance was significantly smaller than the interspecific genetic distance between Euphorbia maculata L. and its adulterants. The phylogenetic tree analysis results showed that the samples of Euphorbia maculata L. were clustered into a single line,which indicated that rbcL and psbA-trnH sequences could distinguish Euphorbia maculata L. and its adulterants.
