籼稻花药培养诱导条件优化

Optimization of induction conditions for anther culture of indica rice

摘要: 以福稻88//华占/甬优2640的F1代为材料进行花药培养。采用正交试验法设置预处理温度、预处理时间、热激温度、噻重氮苯基脲(Thidiazuron,TDZ)浓度4种因素各4个水平,共16种处理组合,对不同条件下花药诱导出愈率进行比较。结果表明,直观分析确定的4个因素最优组合为预处理温度10 ℃、预处理时间8 d、热激温度37 ℃、TDZ浓度0.10 mg/L,但考虑到因素间的交互作用,进一步分析得到最优组合为预处理温度10 ℃、预处理时间8 d、热激温度37 ℃、TDZ浓度0.05 mg/L。

Abstract: Anther culture was carried out with F1 generation of Fudao 88//Huazhan/Yuyou 2640 as material. The orthogonal test method was used to set four levels of pretreatment temperature, pretreatment time, heat shock temperature and concentration of thiazolidine(TDZ); a total of 16 treatment combinations were used to induce anthers under different conditions. The anther induction rate was compared under different conditions. The results show that the optimal combination of the four factors determined by visual analysis ispretreatment temperature 10 ℃, pretreatment time 8 d, heat shock temperature 37 ℃, TDZ concentration 0.10 mg/L. However, considering the interaction between the factors, the optimal combination was obtainedpretreatment temperature 10 ℃, pretreatment time 8 d, heat shock temperature 37 ℃, TDZ concentration 0.05 mg/L.
