乌梅有机酸提取条件优化及其对生物膜形成的抑制作用

Optimization of extraction conditions of organic acids from Prunus mume and its inhibition on biofilm formation

摘要: 对乌梅总有机酸的提取条件进行优化并研究其对金黄色葡萄球菌(Staphylococcus aureus)生物膜产生的抑制作用。通过单因素试验及正交试验对乌梅总有机酸提取条件进行了优化及验证,采用结晶紫染色法对金黄色葡萄球菌生物膜形成量进行了测定。结果表明,乌梅总有机酸最佳提取条件为乙醇70%(体积分数)、料液比3∶40(g∶mL)、超声提取时间1.5 h及超声提取温度50 ℃。在该条件下,乌梅总有机酸提取率为24.50%。提取物浓度为0.5MIC、MIC和2MIC时,对金黄色葡萄球菌生物膜形成的抑制率分别为13.06%、48.24%和74.01%。因此,该提取物对金黄色葡萄球菌生物膜的形成具有抑制作用。

Abstract: The extraction conditions of total organic acid from Prunus mume was optimized and its inhibition on biofilm formation by Staphylococcus aureus was studied. The single factor and orthogonal test were used to optimize and verify the total organic acid extraction scheme of Prunus mume. The amount of biofilm formation by Staphylococcus aureus was determined by crystal violet staining. The results showed that the optimum extraction conditions of total organic acids were ethanol 70% (volume fraction), solid-liquid ratio of 3 g / 40 mL, ultrasonic extraction time of 1.5 h and ultrasonic extraction temperature of 50 ℃. The extraction rate of total organic acid was 24.50% from Prunus mume under these conditions. The inhibitory rates on biofilm formation by Staphylococcus aureus was 13.06%,48.24% and 74.01% respectively at the extract concentration of 0.5MIC, MIC and 2MIC. Therefore, the extract could inhibit the formation of Staphylococcus aureus biofilm.
