石榴3-脱氢奎尼酸合成酶基因的克隆及表达分析

Cloning and Expression Analysis of 3-dehydroquinate Synthase Gene in Pomegranate

摘要: 利用RT-PCR技术从泰山红石榴(Punica granatum L.)果皮中获得3-脱氢奎尼酸合成酶(DHQS)基因cDNA序列,分析了其在不同石榴品种发育期果实中的表达情况。结果表明,DHQS基因cDNA序列长273 bp,编码91个氨基酸,编码蛋白具有典型的DHQ_Fe_ADH超家族保守结构域。该基因核苷酸序列与欧洲山毛榉、梅花和蒺藜苜蓿的相似性分别为91%、87%和88%,氨基酸序列与欧洲山毛榉、苹果、白梨和拟南芥的相似性分别为95%、92%、92%和88%。两个石榴品种发育期果皮中DHQS基因相对表达量逐渐降低,均在7月15日出现峰值。整个发育期,泰山红DHQS基因表达量高于泰山三白甜。

Abstract: The 3-dehydroquinate synthase(DHQS) cDNA sequence was cloned from Taishanhong pomegranate(Punica granatum L.) pericarp with RT-PCR technique. The expression of DHQS gene was analyzed in different pomegranate cultivars fruits during development. The result showed that the cDNA sequence length of DHQS was 273 bp,which encoded 91 amino acids. The protein of DHQS gene contained DHQ_Fe_ADH superfamily conservative structure domain. The similarity of DHQS nucleotide sequence in pomegranate and Fagus sylvatica,Prunus mume,Medicago truncatula were 91%,87% and 88%,respectively. The similarity of DHQS deduced amino acid sequences in pomegranate and Fagus sylvatica,Malus domestica,Pyrus x bretschneideri,Arabidopsis thaliana were 95%,92%,92% and 88%,respectively. The relative expression level of DHQS gene decreased gradually with the fruit development in two pomegranate cultivars,which peak value appeared at July 15th. The relative expression level of DHQS gene in Taishanhong was higher than that in Taishansanbaitian in the whole development period.
