黄曲霉拮抗菌B10-6-1脂肽类抗生素粗提物的性质与抑菌机理研究

Studies on the properties and antibacterial mechanism of crude lipopeptide extracts from Aspergillus flavus antagonistic bacterium Bacillus Amyloliquefaciens B10-6-1

摘要: 采用酸沉醇提法从菌株B10-6-1发酵液中提取脂肽类抗生素粗提物,采用抑菌圈法确定其最低抑菌浓度及抑菌谱,考察温度、pH、蛋白酶、金属离子、EDTA等因素对其抑菌活性的影响;通过孢子萌发法和显微观察法考察脂肽类抗生素粗提物对黄曲霉(Aspergillus flavus)菌孢子萌发和菌丝的作用。结果表明,脂肽类抗生素粗提物的最低抑菌浓度为4 g/L,在40 ℃,pH为8时抑菌活性最高;温度、pH、蛋白酶、金属离子、EDTA对脂肽类抗生素粗提物抑菌活性都没有较明显的影响;该粗提物抑菌谱广泛,对白菜黑斑菌、小麦纹枯菌等9株真菌都有明显抑制作用。脂肽类抗生素粗提物处理菌丝可使菌丝细胞壁出现消融,并使孢子的萌发率降低,即该物质是通过抑制黄曲霉菌孢子萌发和菌丝生长起到抑菌作用的。

Abstract: The crude lipopeptide extract was extracted from the fermentation broth of strain B10-6-1 by acid precipitation. The minimum inhibitory concentration and the antimicrobial spectrum were determined by the inhibition circle method. The effects of temperature, pH, protease, metal ions, EDTA and other factors on the bacteriostatic activity were investigated. The effects of crude lipopeptide extracts on Aspergillus flavus spore germination and hyphae were investigated by spore germination and microscopic observation. The results showed that the minimum inhibitory concentration of crude lipopeptide extract was 4 g/L, with the highest inhibitory activity at 40 ℃ and pH of 8, while temperature, pH, protease, metal ion and EDTA had no obvious effect on the antibacterial activity of crude lipopeptide extract. The antifungal spectrum of the crude extract was extensive and had obvious inhibitory effect on 9 strains of fungi such as Alternaria brassicae and Rhizoctonia cerealis etc. The mycelium treated with lipopeptide extract could dissolve the site of mycelium cell wall and decrease the germination rate of spores, which could inhibit the germination of Aspergillus flavus spores and the growth of hyphae.
