CpSSR analysis of the genetic relationship between Fraxinus hupehensis and “Jingcui” in Jingshan area

Abstract: In order to study the genetic distance and genetic consistency between Fraxinus hupehensis and “Jingcui” at molecular level. The cpSSR (Chloroplast microsatellite) molecular markers technology were applied to determine the genetic relationship between Fraxinus hupehensis and “Jingcui” collected in Jingshan area, then UPGMA clustering map was constructed through combining the data from other species. The results showed that 88 polymorphic bands were amplified by 22 pairs of cpSSR primers, and UPGMA clustering was constructed for 2 samples according to genetic consistency. The 10 species were clustered into 5 categories, the first category included 2 samples, namely “Jingcui” and Leguminous; the second category was composed of Fraxinus hupehensis, Solanaceae, Actinidiaceae, Rosaceae and Myrtaceae; Agaveaceae, Gramineae and Cruciferae were belong to the III, IV, and V category, separately. The results showed that there was a certain affinity between Fraxinus hupehensis and “Jingcui”, but the affinity was not high.
