枯草芽孢杆菌的发酵培养基优化研究

Optimizing on Fermentation Conditions of Bacillus subtilis by Response Surface Analysis Methodology

摘要: 为了得到枯草芽孢杆菌(Bacillus subtilis)发酵的最佳培养基,应用Plackett-Burman设计法对影响枯草芽孢杆菌芽孢形成的关键因子进行筛选,并进一步采用响应面分析法对影响芽孢产量的关键因素进行研究。结果表明,影响枯草芽孢杆菌芽孢产量的关键因素为玉米淀粉和豆粕的浓度。通过响应面分析法的拟合和推算,得到玉米淀粉和豆粕浓度分别为2.48、4.90 g/mL时,此时模型预测发酵最佳产量为105亿/mL,验证值为110亿/mL,预测值与验证值之间吻合较好,比优化前实际产量(80亿/mL)提高了37%。

Abstract: In order to obtain the optimal medium composition for Bacillus subtilis production,the Plackett-Burman design method was used to evaluate the effects of five variables,and the optimum levels of key factors affecting spore production were conducted by response surface analysis methodology. The results showed that,the optimal values of the two parameters determined by response surface analysis methodology were corn starch 2.48 g/mL and corn-soybean 4.90 g/mL. The predicted spore production was 1.05×1010 CFU/mL,and the actual experimental result was 1.10×1010 CFU/mL,they were in agreement and it is 1.37 folds of the production before optimization (8×109 CFU/mL).
