伪狂犬病毒HuB2020株的分离与鉴定

Isolation and identification of pseudorabies virus HuB2020 strain

摘要: 从临床病料中分离纯化1株猪伪狂犬病毒——HuB2020,并对其毒力、培养特性及遗传变异开展了一系列研究。结果表明,该毒株gE基因核苷酸序列与国内PRV毒株的核苷酸序列同源性在98.9%~99.9%。纯化后的病毒经测定其在PK15细胞上的TCID50为107.5/mL;对6周龄昆明小鼠的LD50为1×105.7。组织嗜性分布显示,106.0TCID50/0.1 mL、105.0TCID50/0.1 mL攻毒组对小鼠的神经嗜性均高达100%,且病毒在脑和心脏中的检出率均高于其他组织。

Abstract: A strain of porcine pseudorabies virus:HuB2020 was isolated and purified from clinical materials, and a series of studies were carried out on its virulence, culture characteristics and genetic variation. The results showed thatthe nucleotide sequence of gE gene in this strain was 98.9%~99.9% with that of PRV (Pseudorabies virus,PRV) strain in China. TCID50 of purified virus on PK15 cells was determined to be 107.5/mL. The LD50 of 6-week-old Kunming mice was 1×105.7. Tissue sexual distribution showed that,106.0TCID50/0.1 mL, 105.0 TCID50/0.1 mL challenge groups showed 100% neurotropism to mice,and the detection rate of the virus in brain tissue and heart tissue was higher than other tissues.
