狗蚁草总生物碱对大鼠肝星状细胞胶原蛋白、细胞因子的影响

Effects of total alkaloids from Alysicarpus vaginalis L. DC. on the collagen and cytokines of rat hepatic stellate cells

摘要: 以胶原蛋白、细胞因子为考察对象,探索狗蚁草(Alysicarpus vaginalis L. DC.)总生物碱(AVTA)的抗纤维化作用机制。采用MTT法检测AVTA抑制肝星状细胞增殖的无毒浓度;采用酶联免疫吸附法(ELISA)检测AVTA(10.00、7.00、4.90 μg/mL)对T6细胞上清液 COLⅠ(Ⅰ型胶原)、COLⅢ (Ⅲ型胶原)和PDGF(血小板衍生生长因子)、IGF-1(胰岛素样生长因子-1)、TNF-α(肿瘤坏死因子)及IL-6(白细胞介素-6)含量的影响。结果表明,AVTA抑制肝星状细胞增殖的无毒浓度在13.72 μg/mL以下;与细胞对照组相比,10.00 μg/mL的AVTA能显著抑制COLⅠ、COLⅢ、PDGF、TNF-α、IGF-1、IL-6的分泌(P<0.05或P<0.01)。

Abstract: The anti-fibrosis mechanism of total alkaloids of Alysicarpus vaginalis (AVTA) collagen and cytokines were investigated. MTT assay was used to detect the non-toxic concentration of AVTA inhibiting the proliferation of hepatic stellate cells. And the effect of AVTA(10.00,7.00,4.90 μg/mL) on the content of typeⅠcollagen (COLⅠ), type Ⅲ collagen (COL Ⅲ) and platelet-derived supernatant fluid growth factor (PDGF), tumor necrosis factor (TNF-α), insulin-like growth factor 1 (IGF 1) and Interleukin 6 (IL-6) were detected in T6 cells by ELISA. Results showed that the non-toxic concentration of AVTA inhibiting the proliferation of hepatic stellate cells was less than 13.72 μg/mL; Compared with the cells in the control group, the total alkaloid 10.00 μg/mL could inhibit the secretion of COL Ⅰ, COL Ⅲ, PDGF, TNF-α, IGF-1, IL-6 (P < 0.05 or P < 0.01).
