连香树花芽发育过程中的生理生化特性研究

Study on physiological and biochemical characteristics of Cercidiphyllum japonicum during flower bud development

摘要: 通过检测连香树(Cercidiphyllum japonicum)不同发育时期、不同性别植株花芽中同工酶活性,内源可溶性物质及丙二醛含量变化,在生理生化水平上探究其花芽发育规律和对环境的适应策略。以连香树花芽为材料,采用愈创木酚法、氮蓝四唑法、考马斯亮蓝G-250法、蒽酮法以及2-硫代巴比妥酸(TBA)法分别测定了连香树花芽中的过氧化物酶和超氧化物歧化酶活性、可溶性蛋白质、可溶性糖和丙二醛含量变化。结果表明,连香树花芽发育过程中,过氧化物酶活性先升高后降低,超氧化物歧化酶活性在一定水平上波动,可溶性蛋白质、可溶性糖和丙二醛含量均呈先升高后降低的趋势;雌性花芽超氧化物歧化酶活性、可溶性蛋白质和丙二醛含量总体高于雄性花芽。

Abstract: By detecting the changes of isozyme activity, endogenous soluble substance and malondialdehyde content in flower buds of Cercidiphyllum japonica at different developmental stages and different sexes, the development regularity of flower buds and the adaptation strategy to the environment were explored at the physiological and biochemical level. The activities of peroxidase and superoxide dismutase, the contents of soluble protein, soluble sugar and malondialdehyde in flower buds of Cercidiphyllum japonica were determined by guaiacol method, nitrogen blue tetrazole method, Coomassie Bright Blue G-250 method, anthrone method and 2-thiobarbituric acid (TBA) method, respectively. The results showed that during flower bud development, the activity of peroxidase increased at first and then decreased, the activity of superoxide dismutase fluctuated at a certain level, and the contents of soluble protein, soluble sugar and malondialdehyde increased at first and then decreased; the superoxide dismutase activity, soluble protein and malondialdehyde contents in female flower buds were higher than those in male flower buds.
