恩施地区三种重楼属植物活性成分与抗氧化活性研究

Study on the active components and antioxidant activity of three Paris plants in Enshi

摘要: 对比分析恩施地区3种重楼5年生七叶一枝花[Paris polyphylla Smith var. chinensis (Franch).Hara]、啟良重楼(Paris qiliangiana H.Li, J.Yang & Y.H.Wang)、黑籽重楼(Paris thibetica Franch.)中重楼皂苷Ⅱ活性成分含量,建立气相色谱法(GC)检测方法,检测3种重楼提取物DPPH自由基清除活性,研究其抗氧化能力。乙酸乙酯提取重楼皂苷Ⅱ,采用FID检测器,升温程序为0~3 min,60 ℃,3 ℃/min升至280 ℃,保持20 min;进样口温度310 ℃;载气为氮气(99.999%);流速为1.0 mL/min;进样量为5 μL,不分流。结果表明,GC检测得到重楼皂苷Ⅱ标准品的标准曲线为Y=0.845X+72.18,在浓度512.5~16 400.0 ng/μL范围内线性良好,R2>0.999 5。方法检出限为40.5 ng/μL,定量限为130.6 ng/μL。加标回收率为96.93%~98.33%,RSD为2.72%。GC检测发现七叶一枝花中重楼皂苷Ⅱ含量为0.104 mg/g,黑籽重楼中为0.098 mg/g,啟良重楼中为0.015 mg/g。3种重楼提取物的IC50从强到弱为七叶一枝花(2.372 mg/mL)、啟良重楼(2.461 mg/mL)、黑籽重楼(2.473 mg/mL)。该方法准确可靠,简便快捷且灵敏度高,适用于重楼皂苷Ⅱ含量的测定。恩施地区七叶一枝花中的活性成分重楼皂苷Ⅱ含量最高,其次是黑籽重楼,啟良重楼的含量最低。重楼提取物有一定的抗氧化能力,且七叶一枝花的抗氧化能力最强。

Abstract: The contents of polyphyllin Ⅱ in the five-year-old Paris polyphylla Smith var. chinensis(Franch).Hara, Paris qiliangiana H.Li, J.Yang & Y.H.Wang and Paris thibetica Franch. from three Paris in Enshi were compared and analyzed, and the gas chromatography (GC) was established. The DPPH free radical scavenging activities of three Paris polyphylla extracts were detected and their antioxidant activities were studied. The method for detecting polyphyllin Ⅱ extracted with ethyl acetate was as follows:Using FID detector, the temperature programming was 0~3 min, 60 ℃, the temperature increased to 280 ℃ at 3 ℃/min, and maintained for 20 min; injection port temperature was 310 ℃; the carrier gas was nitrogen (99.999%); the flow rate was 1.0 mL/min; injection volume was 5 μL, no split. The results showed that the standard curve of polyphyllin Ⅱ standard by GC was Y=0.845X+72.18, and the linearity was good within the concentration range of 512.5~164 00.0 ng/μL,with R2> 0.999 5. The limit of detection was 40.5 ng/μL and the limit of quantitation was 130.6 ng/L. The recovery was 96.93%~98.33% with RSD of 2.72%. GC analysis showed that the contents of polyphyllin Ⅱ were 0.104 mg/g, 0.098 mg/g and 0.015 mg/g in P. polyphylla,P. thibetica and P. qiliangiana, respectively. The IC50 values of the three extracts of P. polyphylla,P. qiliangiana, and P. thibetica were 2.372 mg/mL, 2.461 mg/mL and 2.473 mg/mL, respectively, which exhibited strong to weak bacteriostatic activity. The detection method for polyphyllin II was accurate, reliable, simple, sensitive and suitable. The content of the active ingredient polyphyllin Ⅱ in the P. polyphylla in Enshi area was the highest, followed by P. thibetica, and P. qiliangianawas the lowest. The extracts of three Paris plants had certain antioxidant capacity, and P. polyphylla had the strongest antioxidant capacity.
