Effect of maize seed bud size on Agrobacterium transformation

Abstract: The seeds of Zheng 58 maize inbred lines were used as experimental materials. After the corn seeds were sterilized and germinated, the growth points of big and small maize buds were transformed with Agrobacterium. After co-cultivation, the germination was carried out after transformation. By the situation and the results of green fluorescent protein detection of transformed buds, the effect of the size of maize buds on transformation was analyzed, and the size of maize buds suitable for Agrobacterium transformation was determined. The results showed that the maize seed buds were used for Agrobacterium transformation when they were 0.3~0.5 cm in length, which was beneficial to Agrobacterium tumefaciens infection and transformation of maize buds and post-transformation bud regeneration.
