Isolation and identification of pathogen of Lilium davidii var. unicolor scale air culture rot

Abstract: In order to identify the pathogen species of Lilium davidii var.unicolor scale rot during air culture, different diseased tissues were collected, isolated, purified and determined pathogenicity. At the same time, the isolates were identified by morphology and molecular biology based on rDNA-ITS sequence analysis, and the phylogenetic tree was constructed to identify the pathogenic bacteria species. The results showed that a total of 18 fungal strains were isolated and purified, among which 4 strains could cause air culture scale rot of Lilium davidii var. unicolor. The four strains were identified by morphology as Fusarium oxysporum and Fusarium fujikuroi, which belonged to two types of Fusarium. The results of molecular biological identification showed that the four strains clustered in a large tree-like branch with multiple sequences of Fusarium in the phylogenetic tree, among which 1-A-11 and 10-A-2 had more than 99% sequence similarity with Fusarium oxysporum, and strains 10-A-1 and 1-B-1 had more than 99% sequence similarity with Fusarium fujikuroi. It was indicated that Fusarium oxysporum and Fusarium fujikuroi were the pathogens causing the scale rot of Lilium davidii var. unicolor in the process of air culture, and the incidence of Fusarium oxysporum and Fusarium fujikuroi were 95% and 75%, respectively.
