Identification of Cablin from Different Habitats by SCoT Molecular Method

Abstract: : Through the extraction of cablin[Pogostemon cablin (Blanco)Benth.] DNA and orthogonal design of PCR reaction system, screened and obtained cablin SCoT primers by PCR amplification, applied the NTSYS2.10e software and used the UPGMA method for cluster analysis to identify the samples of cablin by molecular perspective method,which was distributed in 16 groups include Guangzhou city,Yangjiang city, Zhaoqing city, Zhanjiang city and Hainan province and so on. Results showed that the SCoT primer sequence amplification of 167 bands, including 108 polymorphism bands, the percentage of polymorphic was 64.7%, and the similarity coefficient between each type distribution between 0.69~0.91, while different regions cablin were respectively divided. It was indicated that cablin has abundant genetic polymorphism, and SCoT molecular marker could be used as one of the basis for the identification of cablin.
