铁十字酢浆草不同极性部位的HPLC指纹图谱与抗氧化活性研究

Study on antioxidant activity and HPLC fingerprints from different polarity parts of Oxalis corniculata L.

摘要: 分别采用酒石酸亚铁显色法、亚硝酸钠-硝酸铝-氢氧化钠显色法、硫酸-苯酚法与DNS法,测定酢浆草(Oxalis corniculata L.)鳞茎外皮和内芯中多酚、黄酮、多糖的含量。以乙腈和0.1% 磷酸水溶液作流动相,梯度洗脱,考察铁十字酢浆草醇提液不同极性部位的HPLC指纹图谱;以抗坏血酸为阳性对照,采用1,1-二苯基-2-苦肼基(DPPH)自由基清除法、羟基(·OH)自由基清除法测定体外抗氧化活性。鳞茎外皮和内芯中多酚含量分别为5.45、4.73 mg/g,黄酮含量分别为16.62、7.70 mg/g,多糖含量分别为17.00、121.02 mg/g。在280 nm波长下,以21号峰为基准峰标定醇提液外皮共有峰32个,内芯31个。不同极性部位的共有峰为外皮:石油醚相8个,氯仿相22个,乙酸乙酯相22个,正丁醇相26个,水相9个;内芯:石油醚相14个,氯仿相21个,乙酸乙酯相17个,正丁醇相19个,水相10个。在抗氧化活性测定体系中,鳞茎外皮和内芯在不同极性部位均有一定的抗氧化活性。氯仿相、乙酸乙酯相对两种自由基都表现出较好的抗氧化活性。铁十字酢浆草鳞茎中富含多酚、黄酮、多糖等成分,醇提物经石油醚、氯仿、乙酸乙酯、正丁醇依次萃取后,各分离相化学指纹图谱和抗氧化能力均存在差别,可为后期药效物质发现提供参考。

Abstract: The contents of polyphenol, flavonoids, polysaccharide in the bulb of Oxalis corniculata L. were measured by ferrous tartrate method, erinitrit-aluminum nitrate-sodium hydroxide method, phenol-sulfuric acid method and DNS method. Using acetonitrile and 0.1% phosphoric acid as mobile phase and gradient elution, the fingerprints of different polar parts of alcohol extract of Oxalis ferruginosa were investigated. The antioxidant activity in vitro was determined by 1,1-diphenyl-2-picrylhydrazine radical scavenging method and hydroxyl. OH radical scavenging method with ascorbic acid as positive control. The polyphenol contents of the two parts were 5.45 and 4.73 mg/g. The flavonoids contents of the two parts were 16.62 and 7.70 mg/g. The polysaccharide contents of the two parts were 17.00 and 121.02 mg/g. 32 peaks were calibrated on the basis of peak 21, and 31 inner cores were calibrated were set at 280 nm. The common peaks in different polar parts are the outer skin: petroleum ether phase 8, chloroform phase 22, ethyl acetate phase 22, n-butanol phase 26, water phase 9; inner core: petroleum ether phase 14, chloroform phase 21, ethyl acetate phase 17, n-butanol phase 19, water phase 10. In the determination system of antioxidant activity, the outer skin and inner core of bulb had certain antioxidant activity in different polar parts. The chloroform phase and ethyl acetate showed better antioxidant activity than the two free radicals. The contents of polyphenol, flavonoids and polysaccharide were rich in the bulb of Oxalis corniculata L.. After extraction by petroleum ether, chloroform, ethyl acetate and n-butanol, the chemical fingerprints and antioxidant ability of the ethanol extracts were different, which could provide reference for the discovery of effective substances in the later period.
