甘蓝型油菜花粉粒ARTP诱变及后代表型鉴定

ARTP irradiation on pollen grain of Brassica napus and phenotype identification of their offsprings

摘要: 为获得高含油量油菜的变异株系,开展了甘蓝型油菜(Brassica napus L.)花粉粒ARTP诱变试验。以甘蓝型油菜Ogu-CMS恢复系16C花粉为诱变材料,采用ARTP诱变技术分别辐照0、20、30、40 min,再将诱变后的花粉授于同一群体中自然产生的不育单株16CS。调查不同ARTP辐照时间下的诱变花粉授粉当代单角粒数及后代子粒发芽势、发芽率、不育株率和含油量的变化情况。结果表明,ARTP辐照甘蓝型油菜花粉30 min 以上,能显著降低诱变花粉授粉当代单角粒数及后代M0代子粒的发芽势和发芽率,提升M0代植株的不育株率,提升M1代子粒的含油量,获得了含油量高达52.61%突变株系ARTP30-315。初步表明利用ARTP-P仪开展油菜花粉粒诱变是有效的,较为适宜的ARTP辐照时间为30~40 min。为生产上应用ARTP-P仪开展甘蓝型油菜花粉粒诱变探索出一个较为适宜的参数体系,为开展油菜种质创新提供了新的方法与思路。

Abstract: In order to obtain the mutant lines of high oil content rape, the ARTP mutation experiment of pollen grains of Brassica napus was carried out.The pollen of the restoring line 16C of Ogu CMS in Brassica napus was irradiated with ARTP mutation technology for 0, 20, 30 and 40 min respectively, and the mutated pollen was given to the sterile single plant 16CS naturally produced in the same population. Then traits of the progeny, such as seed number per pod, seed germination rate and energy and oil content were surveryed. The results showed that ARTP irradiation of Brassica napus pollen for more than 30 min could significantly reduce the number of single horned grains at the time of mutagenic pollen pollination and the germination potential and germination rate of M0 generation seeds, improve the sterile plant rate of M0 generation plants and the oil content of M1 Generation seeds, and obtain the mutant line ARTP30-315 with an oil content of 52.61%. The preliminary results showed that it was effective to carry out rape pollen mutation by ARTP-P instrument, and the more suitable ARTP irradiation time was 30~40 min. A more suitable parameter system was explored for carrying out pollen mutation of Brassica napus by ARTP-P instrument in production, which provided a new method and idea for carrying out germplasm innovation of Brassica napus in the future.
