高效液相色谱-荧光检测法测定大白菜生素K1含量

Determination of vitamin K1 content in Chinese cabbage by high performance liquid chromatography-fluorescence detection

摘要: 为准确测定样品中维生素K1的含量,建立了基于高效液相色谱-荧光检测的大白菜(Brassica rapa L. ssp. pekinensis)维生素K1高灵敏检测技术。采用经优化的流动相,正己烷涡旋提取待测样品,通过中性氧化铝柱净化后使用C18反相色谱柱对试样进行定量分析。结果表明,在浓度为0.229~6.197 μg/mL时维生素K1线性关系良好,相关系数(R2)为0.996 9。此方法检出限为0.29 μg/100 g,定量限为0.95 μg/100 g,回收率为97.5%~101.8%。该方法出峰时间早、能有效分离杂峰、检出限高,是对大白菜中维生素K1进行检测的优选方法。

Abstract: In order to accurately determine the content of vitamin K1 in samples, a highly sensitive detection technology for vitamin K1 in Chinese cabbage (Brassica rapa L. ssp. pekinensis) based on high performance liquid chromatography-fluorescence detection was established. The optimized mobile phase and hexane vortex were used to extract the samples to be tested. After purification by the neutral alumina column, the samples were quantitatively analyzed by the C18 reverse-phase chromatography column. The results showed that vitamin K1 had a good linear relationship within the range of 0.229~6.197 μg/mL, and the correlation coefficient(R2) was 0.996 9. The detection limit of this method was 0.29 μg/100 g, and the limit of quantification was 0.95 μg/100 g. Recovery rate was 97.5%~101.8%. This method had the advantages of early peak time, effective separation of miscellaneous peaks and high detection limit, which was the optimal method for the detection of vitamin K1 in Chinese cabbage.
