球孢白僵菌利用D-PPA合成D-HPPA酵条件优化

Optimization of fermentation conditions for the synthesis of D-HPPA from D-PPA by Beauveria bassiana

摘要: 为了提高球孢白僵菌(Beauveria bassiana)利用R-(+)-2-苯氧基丙酸(D-PPA)合成R-(+)-2-(4-羟基苯氧基)丙酸(D-HPPA)的能力,在单因素试验的基础上,通过Plackett-Burman设计和Box-Behnken中心组合设计优化球孢白僵菌生物合成D-HPPA的液态发酵条件。结果表明,当D-PPA初始添加浓度为40 g/L时,液态发酵最优条件为葡萄糖38.70 g/L、蚕蛹粉15 g/L、FeCl2 1.0 g/L、接种量15%(V/V)、pH 6.6及发酵温度28 ℃时,D-HPPA的产量为(25.10±0.43) g/L,D-HPPA的产率为57.24%±0.97%,比优化前提高了159%。

Abstract: In order to improve the ability of Beauveria bassiana to synthesize R-(+)-2-(4-hydroxyphenoxy) propionic acid (D-HPPA) from R-(+)-2-phenoxy propionic acid (D-HPPA), on the basis of single factor test, the liquid fermentation conditions of D-HPPA biosynthesis by Beauveria bassiana were optimized by Plackett-Burman design and Box-Behnken center combination design. The results showed that when the initial concentration of D-PPA was 40 g/L, the optimal conditions for liquid fermentation were glucose 38.70 g/L, chrysalis powder 15 g/L, FeCl2 1.0 g/L, inoculation amount 15% (V/V), pH 6.6 and fermentation temperature 28 ℃. The yield of D-HPPA was (25.10±0.43) g/L, and the yield of D-HPPA was 57.24%±0.97%, which was 159% higher than that before optimization.
