葡萄叶多酚提取工艺优化及其体外抗氧化、降糖活性研究

Optimization of extraction of polyphenols from grapevine (Vitis vinifera L.) leaf and their in vitro antioxidant and hypoglycemic activities

摘要: 旨在建立并优化超声辅助乙醇提取葡萄(Vitis vinifera L.)叶多酚工艺,并评价纯化多酚组分的体外抗氧化与降糖活性。以葡萄叶为材料,采用超声辅助乙醇提取葡萄叶多酚,采用单因素结合Box-Behnken响应面法优化葡萄叶多酚提取参数,并评价其对多酚提取量的影响。结果表明,在超声温度69 ℃、超声时间51 min、乙醇体积分数61%、液料比40∶1(mL∶g)的优化条件下,葡萄叶粗提液多酚提取量达(36.56±0.86)mg/g。经大孔树脂纯化后,在体外试验中,葡萄叶多酚表现出较强的抗氧化活性,对DPPH和ABTS+自由基清除能力的IC50 分别为0.037 3 mg/mL和0.091 3 mg/mL。同时,葡萄叶多酚对α-淀粉酶和α-葡萄糖苷酶具有显著抑制作用,其IC50分别为0.076 3 mg/mL和0.147 0 mg/mL。表明葡萄叶多酚具有良好的抗氧化和降糖潜力,为葡萄叶农业副产物的资源化利用提供参考。

Abstract: This study aimed to establish and optimize an ultrasound-assisted ethanol extraction process for grapevine (Vitis vinifera L.) leaf, and to evaluate the in vitro antioxidant and hypoglycemic activities of the purified polyphenol fractions. Taking grapevine leaf as the material, the ultrasonic-assisted extraction method was applied to extract polyphenols. Single-factor experiments combined with a Box-Behnken response surface methodology were employed to optimize the extraction parameters and evaluate their effects on polyphenol yield. The results showed that the optimal extraction technology was as follows: ultrasonic temperature of 69 °C, ultrasonic time of 51 min, ethanol volume fraction of 61%, and liquid to material ratio of 40∶1 (mL∶g). The yield of polyphenols in the crude grapevine leaf extract reached (36.56±0.86) mg/g. After purification with macroporous resin, grapevine leaf polyphenols exhibited strong antioxidant activity in vitro, with IC50 values for DPPH and ABTS+ radical scavenging of 0.037 3 mg/mL and 0.091 3 mg/mL, respectively. Meanwhile, the grapevine leaf polyphenols showed significant inhibitory effects against α-amylase and α-glucosidase, with IC50 values of 0.076 3 mg/mL and 0.147 0 mg/mL, respectively. These results indicated that grapevine leaf polyphenols possessed strong antioxidant and hypoglycemic potential, providing a reference for the resource utilization of grapevine leaf agricultural by-products.
