Abstract: In order to obtain the optimal medium composition for Trichoder mareesei production, the Plackett-Burman design was used to evaluate the effects of variables, and the optimum levels of key factors affecting mycelium yield were conducted by response surface analysis methodology. The results showed that, the optimal values of the two parameters determined by response surface analysis methodology were fish peptone 1.414 3 g/100 mL and glycerol 2.439 3 g/100 mL. At this point, the model predicted optimal fermentation yield was 85 g/100 mL, and the verified value was 81 g/100 mL,and they were in agreement and it was about 1.35 times the yield of the original medium (60 g/100 mL).
