Optimizing on Fermentation Conditions of Bacillus subtilis by Response Surface Analysis Methodology

Abstract: In order to obtain the optimal medium composition for Bacillus subtilis production,the Plackett-Burman design method was used to evaluate the effects of five variables,and the optimum levels of key factors affecting spore production were conducted by response surface analysis methodology. The results showed that,the optimal values of the two parameters determined by response surface analysis methodology were corn starch 2.48 g/mL and corn-soybean 4.90 g/mL. The predicted spore production was 1.05×1010 CFU/mL,and the actual experimental result was 1.10×1010 CFU/mL,they were in agreement and it is 1.37 folds of the production before optimization (8×109 CFU/mL).
