Comparision of physiological indexes of the first leaf and second leaf in the growth of soybean seedlings

Abstract: Soluble protein content, photosynthetic pigment content, MDA content, ascorbic acid content, enzyme activity and peroxidase activity of hydrogen peroxide in the first leaf and the second leaf of soybean [Giycine max (Linn.) Merrill] seedlings were determined. The results showed that there was no significant difference in soluble protein content between the first and second true leaf. The content of chlorophyll a and carotenoid in the second leaf was 23.4% and 89.9% higher than those in the first leaf, respectively. The content of chlorophyll b in the first leaf was 76.9% higher than that in the second leaf. The total photosynthetic pigment content in first leaf and second leaf were close to each other. The value of chlorophyll a/b in the second leaf was about 2.18 times higher than that in the first true leaf. The MDA content in the first true leaf was 3.04 times higher than that in the second true leaf. The content of ascorbic acid in the second leaf was 1.52 times higher than that in the first true leaf. The peroxidase activity and catalase activity could not been determined in these two leaves. Besides that, ascorbic acid content was negatively correlated with the content soluble protein, MDA and chlorophyll b, and was positively correlated with the content of chlorophyll a and carotenoid.
