Study on high efficient transformation of exogenous fatty acids into hydroxyfatty acids by engineering Escherichia coli

Abstract: Cytochrome P450BM3 can hydroxylates fatty acids into hydroxyfatty acids(HFAs) at the ω-positions with high entioselectivity. Based on the construction of Escherichia coli carrying P450BM3 gene to transform fatty acids. As an effort to solve inhibit fatty acid β-oxidation and increase outer membrane fatty acids transporter, the lipoacyl-coa synthase gene(fadD) deletion and outer membrane proteins encode gene(fadL) overexpression strains were built. Under shake-flask conditions, this recombinant strains produced 748.8 mg/L hydroxy dodecanoic acid, meanwhile the conversion rate was 93.6%.
