Effects of total alkaloids from Alysicarpus vaginalis L. DC. on the collagen and cytokines of rat hepatic stellate cells

Abstract: The anti-fibrosis mechanism of total alkaloids of Alysicarpus vaginalis (AVTA) collagen and cytokines were investigated. MTT assay was used to detect the non-toxic concentration of AVTA inhibiting the proliferation of hepatic stellate cells. And the effect of AVTA(10.00,7.00,4.90 μg/mL) on the content of typeⅠcollagen (COLⅠ), type Ⅲ collagen (COL Ⅲ) and platelet-derived supernatant fluid growth factor (PDGF), tumor necrosis factor (TNF-α), insulin-like growth factor 1 (IGF 1) and Interleukin 6 (IL-6) were detected in T6 cells by ELISA. Results showed that the non-toxic concentration of AVTA inhibiting the proliferation of hepatic stellate cells was less than 13.72 μg/mL; Compared with the cells in the control group, the total alkaloid 10.00 μg/mL could inhibit the secretion of COL Ⅰ, COL Ⅲ, PDGF, TNF-α, IGF-1, IL-6 (P < 0.05 or P < 0.01).
