Abstract: To determine the optimal process conditions for ultrasonic extraction of polyphenols from Pteris semipinnata L. and to improve the extraction yield, response surface methodology was used to optimize the extraction process. On the basis of single-factor experiments, five factors including solid-liquid ratio, ethanol concentration, extraction time, extraction temperature and mesh number were selected, and the extraction yield of polyphenols from Pteris semipinnata was taken as the response value; the effects of each factor and their interactions on extraction efficiency were analyzed, and the reliability of the optimized results was confirmed by validation experiments. The results showed that the order of influence of each factor on polyphenol extraction yield from high to low was extraction temperature, solid-liquid ratio, extraction time, mesh number, and ethanol concentration; the optimal process conditions were solid-liquid ratio of 1∶50, ethanol concentration of 60%, extraction time of 53 min, extraction temperature of 53 ℃, and mesh number of 160, under which the polyphenol extraction yield was 25.94 mg/g, close to the model-predicted value (26.12 mg/g). This ultrasonic extraction process was stable and feasible, and the optimization effect was significant, which could provide a methodological reference for the optimization of extraction processes of polyphenols from similar plants.
