Idengtification and Lcation of the Selenium Resistant Gene in Arabidopsis thaliana

Abstract: A selenate-torlerance arabidopsis mutant was obtained by using the root-bending trait under selenate stress. It was conformed that the selenate resistant characteristics could be steady inheritance. Then this mutant was crossed with the wild-type Ler. and F1 generation were created with heterozygous type completely. The F2 crossed by theirself were expressed the separation ratio of 3∶1 in response of sensitive selenium to resistent selenium,It were suggested that this mutant was a monogenic recessive mutation. Map-based cloning population were composed of the 132 selenate-tolerance seedlings in F2 generation. Using SSLP and In/Del based on PCR to preliminary chromosomal location. Through linkage analysis,it's indicated that this mutant gene was located on the chromosome I,which was mapped between molecular marker F12P19 and NGA111.
