The Optimization of Acetylcholinesterase-Spectrophotometry for Determination of Organophosphate and Carbamate Pesticide Residues in Vegetables

Abstract: Acetylcholinesterase-spectrophotometry is the most widely used method for rapid detection of organophosphorus and carbamate pesticide residues in vegetables. However,in practical application,the method is seriously limited by numerous peripheral influencing factors and low accuracy. The optimization of Acetylcholinesterase-Spectrophotometry for determination of organophosphate and carbamate pesticide residues in vegetables were studied in this paper.Controlling the ΔA value between 0.2~0.3 and its results were given,which could improve the accuracy of positive test results. Extra thermostatic bath was chosen instead of device came with instrument and the results showed greatly improved reproducibility and parallelism. In order to avoid the influence of acetone on enzyme activity,the added amount of acetone should be controlled less than 1% when preparing standard solution.
