Study on pollen germination and pollen tube growth culture in vitro of Michelia figo

Abstract: To clear the influence of boric acid, calcium, sucrose, PEG-4000 and pH on pollen germination and pollen tube growth culture in vitro of Michelia figo, the pollen of Michelia figo was studied by liquid culture. The results showed that boric acid, calcium, sucrose, PEG-4000 and pH in the medium had a significant influence on pollen germination and pollen tube growth of Michelia figo. The optimal concentration of boric acid was 0.03 g/L, calcium was 0.07 g/L, sucrose was 100 g/L, PEG-4000 was 150 g/L, and pH was 7.0. The pollen germination rate was 81.74%, and the length of the pollen tube was 424.17μm. In this study, the optimal medium concentration of pollen germination and pollen tube growth in vitro culture of Michelia figo was obtained, which provided a theoretical basis for the conservation of its germplasm resources and the breeding of new varieties.
