The effect of four kinds of medium types on Verticillium dahliae V991b and its mutant

Abstract: In order to understand the biological characteristics and pathogenicity of Verticillium dahilae V991b and its mutant V991w, the effects of different conditioned medium on the growth and pathogenicity of the two pathogens were investigated. The results showed that the growth curve of two strains on different types of medium was linear basically. The growth rate of V991b on PDA was significantly different from other types of medium. And the growth rate of V991w on PDA and PCA medium was significantly different from other two types of medium. After cultiviated 15 days on PDA medium, there was no significantly difference between the colony diameter of V991b and that of V991w. During amplified the fungi in Czapek’s liquid medium and then inoculated the cotton seedling, the pathogenicity of V991b and V991w was quite different. The disease index on 25 days of inoculated was 56.63 and 23.23, respectively. However, the disease index of cotton seedlings inoculated with Czapek’s-cotton leaf tissue liquid medium was significantly reduced, and the disease index of V991b and V991w at 25 d inoculation was only 12.12 and 7.32. The plant tissue culture medium had no significant effect on the diameter of the microsclerotia, but it had significant inhibitory effect on the number of microsclerotia and the growth of hypha.
