酶法辅助提取苦皮藤素工艺优化

Optimization of enzyme assisted extraction of celangulin from Celastrus angulatus

摘要: 以苦皮藤素(Celastrus angulatus Maxim)根皮为原料,以苦皮藤素得率为指标,对碱性蛋白酶、纤维素酶、胃蛋白酶、中性蛋白酶4种酶辅助提取苦皮藤素的效果进行比较分析,筛选出酶解效果较好的胃蛋白酶,再通过单因素试验和正交试验,优化胃蛋白酶辅助提取苦皮藤素的最佳工艺。结果表明,胃蛋白酶辅助提取苦皮藤素在酶解温度37 ℃、时间5 h、pH 2.0、加酶量66 000 U/g的条件下效果最好。

Abstract: Celangulin from the root skin of Celastrus angulatus were isolated using an enzye-assisted procedure. By analyzed the effects of alkaline protease, cellulase, pepsin and neutral protease on the extraction of celangulin, Pepsase was selected to optimize the extraction conditions. The results showed that the optimal conditions for celangulin extraction was found to be extraction with enzymatic hydrolysis with 66 000 U/g pepsase at 37 ℃ and pH 2 for 5 h. Under these conditions, the maximum yield of celangulin was obtained.
