凸尖杜鹃组培快繁体系的构建

Establishment of the tissue culture and rapid propagation system on Rhododendron sinogrande Balf. f. et W. W. Sm.

摘要: 通过对凸尖杜鹃(Rhododendron sinogrande Balf. f. et W. W. Sm.)实生苗的启动与增殖培养,探索适宜凸尖杜鹃的灭菌方法及其启动与增殖培养基配方,以建立凸尖杜鹃的组织培养体系。结果表明,两种杜鹃用0.1%的HgCl2灭菌8 min真菌污染率较低,在此处理下凸尖杜鹃和泡泡叶杜鹃(Rhododendron edgeworthii Hook. f.)的真菌污染率分别为5%和10%;当培养基中的吲哚丁酸(IBA)浓度为0.3 mg/L、玉米素(ZT)浓度为0.01 mg/L时,凸尖杜鹃实生苗的启动率最高,达到80%;当噻苯隆(TDZ)的浓度为 6 mg/L时,增殖倍数最高,达到5.32。

Abstract: Through the initiating and proliferating culture of Rhododendron sinogrande seedling, the sterilization method and media components of initiating and proliferating culture suitable for R. sinogrande were explored to establish the tissue culture system of R. sinogrande. The results showed that, the fungus contamination rate of two kinds of Rhododendron were lower when they were sterilized for eight minutes under 0.1% HgCl2, and the fungus contamination rate for R. sinogrande and R. edgeworthii Hook. f. was 5% and 10%, respectively. The initiation rate of R. sinogrande seeding was the highest for 80%, when the concentration of IBA was 0.3 mg/L and the concentration of ZT was 0.01 mg/L in the medium; The multiple proliferation was the highest for 5.32, when the concentration of TDZ was 6 mg/L in the medium.
