凤头姜组培快繁体系研究

Study on tissue culture and rapid propagation of Zingiber officinale Roscoe cv. Fengtoujiang

摘要: 利用组培技术,以凤头姜(Zingiber officinale Roscoe cv.Fengtoujiang)茎尖为外植体,采用不同激素组合,筛选最佳培养基配方,构建凤头姜脱毒快繁体系。结果表明,MS+6-BA(1.5 mg/L)+NAA(0.1 mg/L)培养基对愈伤组织及茎尖成活率的影响最佳;MS+6-BA(2.0 mg/L)+KT(0.1 mg/L)培养基诱导愈伤组织形成的丛生芽多,且粗壮。MS+6-BA(1.0 mg/L)+NAA(0.3 mg/L)培养基不仅对根的分化最有效,还能促进分蘖及成苗,大大提高了繁殖系数。

Abstract: To screen out the optimal culture medium and build the virus-free rapid propagation system for Fengtoujiang,experiment was carried out with tissue culture technology,using stem apex of as explant and different phytohormone combination.The results indicated that the optimal effect on survival of the callus and stem apex was the combination of MS+6-BA(1.5 mg/L)+NAA(0.1 mg/L).The formation of callus was induced by the medium of MS+6-BA(2.0 mg/L)+KT(0.1 mg/L)and the robust adventitious bud was increased as well.Not only had the MS+6-BA(1.0 mg/L)+NAA(0.3 mg/L) the most efficient effect on ginger root differentiation,but also improved tillering and stand establishment,which increased the propagation significantly.
