栝楼组织培养育苗技术研究

Study on tissue culture seedling technology of Trichosanthes kirilowii Maxim.

摘要: 以栝楼(Trichosanthes kirilowii Maxim.)当年新长出的嫩梢作为外植体,成功建立了栝楼组织培养育苗技术体系。结果表明,外植体最适合的消毒方法为先使用75%的乙醇消毒20 s,然后使用0.1% HgCl2消毒4 min;最佳诱导培养基配方为MS+2.0 mg/L KT+0.4 mg/L IAA,最佳增殖培养基配方为MS+3.0 mg/L KT+0.4 mg/L IAA,最佳生根培养基配方为1/4 MS+0.2 mg/L NAA;移栽的最佳种植基质体积配比为珍珠岩∶草炭∶蛭石=1∶2∶1。

Abstract: The tissue culture system of Trichosanthes kirilowii Maxim.was successfully established by using the young shoots as explants. The results showed that the most suitable disinfection method for explants was to use 75% ethanol for 20 s and then 0.1% HgCl2 for 4 min. The best induction medium was MS+2.0 mg/L KT+0.4 mg/L IAA, the best proliferation medium was MS+3.0 mg/L KT+0.4 mg/L IAA, and the best rooting medium was 1/4 MS+0.2 mg/L NAA. The best planting substrate volume ratio for transplanting was perlite ∶ peat ∶ vermiculite = 1∶2∶1.
