Changes of endogenous growth regulators during roots of tissue culture seedlings of Lonicera macranthoides Hand.-Mazz

摘要: 以灰毡毛忍冬(Lonicera macranthoides Hand.-Mazz)组培苗为试验材料,用双抗体一步夹心酶联免疫法测定灰毡毛忍冬生根诱导过程(0、6、12、18、24、30 d)中生长素(IAA)、脱落酸(ABA)、赤霉素(GA3)、玉米素核苷(ZR)含量的变化。结果表明,ABA含量呈先上升后下降的趋势;IAA、GA3和ZR含量均呈先下降后上升的趋势。IAA、GA3含量和IAA/ABA比值与生根率呈显著或极显著正相关,ABA含量与生根率呈负相关,ZR含量与生根率相关性不显著。

Abstract: The tissue culture seedlings of Lonicera macranthoides Hand.-Mazz were used as experimental materials,the changes of the content of indole-3-acetic acid (IAA), abscisic acid (ABA), gibberellic acid (GA3), and zeatin nucleoside (ZR) in the root-inducing process (0,6,12,18,24,30 d) were determined by the method of enzyme-linked immunosorbent assay(ELISA). The results showed that the content of ABA was increased initially and then decreased. The concentrations of IAA, GA3 and ZR displayed the opposite trends, which were decreased firstly and subsequently increased. The contents of IAA and GA3 and the ratio of IAA to ABA were significant or extremely significant positively correlated with rooting rates, while ABA contents were negatively correlated with rooting rates, and there was no significant correlation between ZR content and rooting rate.
