PAM细胞中猪MMP14不同转录本检测及其生物信息学分析

Identification of different transcripts of porcine MMP14 in PAM cells and thier bioinformatics analysis

摘要: 基质金属蛋白酶14(MMP14)是基质金属蛋白酶家族中首个被发现的膜型基质金属蛋白酶。MMP14蛋白能水解多种细胞外基质组分和血清蛋白,促进细胞外基质降解,并可水解细胞因子、趋化因子和生长因子等。研究发现猪肺泡巨噬细胞(PAM)中存在2种猪MMP14转录本,利用荧光定量PCR技术检测了PRRSV感染的猪PAM细胞中猪MMP14基因不同转录本的表达水平变化,发现PRRSV感染后导致转录本NM_214239.2显著下调表达(P<0.05),而转录本XM_021098303.1极显著上调表达(P<0.01)。通过生物信息学分析发现,2种转录本编码的蛋白前肽区序列存在明显差异。XM_021098303.1编码的蛋白比NM_214239.2编码的蛋白序缺少了RRKR基序和PG_binding_1功能区,可能导致MMP14蛋白不具有功能活性。推测PRRSV感染后抑制MMP14活性,与PRRSV刺激细胞因子与趋化因子的大量表达相关。然而,MMP14在PRRSV感染中的具体作用还有待进一步研究。

Abstract: Matrix metalloproteinases 14 (MMP14) is the first discovered membrane-type matrix metalloproteinases in the matrix metalloproteinases super family.MMP14 protein can hydrolyze various extracellular matrix components and serum proteins,promote extracellular matrix degradation,and hydrolyze cytokines,chemokines and growth factors.In this study,two different porcine MMP14 transcripts were found in pig alveolar macrophages (PAM).The expression levels of different transcripts of MMP14 gene in porcine PAM cells infected with PRRSV were detected by fluorescence quantitative PCR,the results showed that the expression level of transcript NM_214239.2 significantly reduced (P<0.05),while XM_021098303.1 significantly incresed (P<0.01) in PRRSV infected PAMs.Bioinformatics analysis results showed that these two different transcripts encoded two different proteins.Compared with NM_214239.2 encoded protein,the RRKR motif and PG_binding_1 functional region were missing in XM_021098303.1 encoded protein,which might not havefunctionalactivity in the inactivation of MMP14 protein.It is speculated that the inactivation of MMP14 after PRRSV infection maybe relate to the high expression of cytokines and chemokines stimulated by PRRSV.However,the specific role of MMP14 in PRRSV infection still needs further study.
