HPLC法同时测定养心草中五种成分的含量

Determination of 5 chemical constituents in Sedum aizoon L. by HPLC

摘要: 建立HPLC法同时测定养心草中5种成分的含量并采用单因素和正交试验筛选最优提取工艺。结果表明,采用建立的HPLC含量测定方法可以同时准确测定5种成分的含量,杨梅苷、芦丁、槲皮苷、槲皮素、山柰酚分别在22~88 μg/mL、5~20 μg/mL、18~64 μg/mL、0.32~1.28 μg/mL、0.56~2.24 μg/mL,与峰面积线性关系良好,加样回收率在95.61%~107.38%,回收率的RSD为1.37%~4.60%,均符合要求,最优提取工艺为50%甲醇90 ℃加热回流提取30 min。

Abstract: To establish a HPLC method to simultaneously determine the content of 5 compounds in Sedum aizoon L. and use single factor and orthogonal experiments to select the optimal extraction process. The results showed that the established HPLC method can be used to determine the content of five components accurately at the same time, the concentrations of myricetin, rutin, quercitrin, quercetin and kaempferol in 22~88 μg/mL, 5~20 μg/mL, 18~64 μg/mL, 0.32~1.28 μg/mL, 0.56~2.24 μg/mL have a good linear relationship with the peak area, respectively; the range of recovery rate of 5 compounds was 95.61%~107.38%; and the RSD values was 1.37%~4.60%. The optimal extraction process was 50% methanol, 90 ℃ heating reflux extraction for 30 min.
