三种地生兰根际真菌的分离与检测

Isolation and identification of three terrestrial orchids rhizosphere fungi

摘要: 选取兰属的春兰、春剑和寒兰3种地生兰,对其栽培基质中的根际真菌进行分离培养后得到3个优势菌株,并以18SDNA序列扩增鉴定为米根霉(Rhizopus oryaze)、球毛壳菌(Chaetomium lobasum)、小孢根霉(Rhizopus icrosporus),通过对兰花根系进行染色显微观察,发现兰花根与真菌共生形成的菌根结构,为兰科植物根际真菌研究提供了理论依据。

Abstract: Three Chinese terrestrial orchids Cymbidium goeringii, Cymbidium tortisepalum Fukuy.var. longibracteatum and Cymbidium kanran were chosen to the samples for isolating and culturing rhizosphere fungi from substrates, which found and identificated three preponderant fungal strains during the storage, and which identified as Rhizopus oryaze, Chaetomium lobasum, Rhizopus icrosporus by 18SDNA sequences. It was found that the roots of orchid were mutualized with fungi to form rhizosphere structure, according to orchid roots stained and microscopical observed, so as to provide a theoretical basis for further research of orchids rhizosphere fungi.
