Study on tissue culture technology of Dendrobium candidum Wall. ex Lindl. cv. Heijie

Abstract: The protocorm induction and proliferation as well as seedling’ rooting of Dendrobium candidum Wall. ex Lindl. cv. Heijie stem tip were studied preliminarily. Dendrobium candidum Wall. ex Lindl. cv. Heijie stem tip was sterilized with 75% alcohol for 30 s, and then the epidermis was removed. The best protocorm induction and proliferation medium of Dendrobium candidum Wall. exLindl. cv. Heijie stem tip was 1/2 MS+5.5 g/L agar+0.55 mg/L 6-Benzylaminopurine+0.55 mg/L 1-Naphthalenea acid+0.1 mg/L AgNO3+100 g/L mashed potato+20 g/L white granulated sugar+0.45 g/L activated carbon. The best rooting medium of Dendrobium candidum Wall. exLindl. cv. Heijie seedling was 1/2 MS+5.5 g/L agar+0.15 mg/L 6-Benzylaminopurine+0.55 mg/L 1-Naphthalenea acid+0.1 mg/L AgNO3+100 g/L banana puree+20 g/L white granulated sugar+0.25 g/L activated carbon.
