Abstract: Using pot experiments, the effects of different concentrations paclobutrazol (PP333) on the growth and physiological changes of Sophora japonica ‘Jinhuai’ seedlings were studied. The results showed that, with the increase of PP333 concentration, tree height, leaf area and taproot length of Sophora japonica ‘Jinhuai’showed a declining trend, whereas base diameter, leaf length to width ratio, fresh weight of root, and root-shoot ratio showed an increasing trend, and the main root diameter gradually increases within the range of 300~500 mg/L. The effects of PP333 treatment on the physiological indexes of ‘Jinhuai’ is: chlorophyll content increases first and then decreases, reaching the maximum value at 375 mg/L, relative water content, soluble sugar and soluble protein content in leaves were effectively increased, MDA content and relative conductivity were effectively reduced compared with that in the control, the activities of POD and CAT reached the maximum when the concentration of paclobutrazol was 500 mg/kg and 750 mg/L, and the activity of SOD increased with the concentration of paclobutrazol; ‘Jinhuai’ seedlings had the strongest resistance with 750~ 1 500 mg/L PP333. PP333 can improve its resistance by regulating the growth and physiological processes of ‘Jinhuai’ and 750 mg/L was best.
