Preliminary Study on Rapid Propagation in Vitro of Three Kinds of Lactuca sativa

Abstract: In this experiment,three kinds of cotyledons from Lactuca sativa were used as explants to study the effects of different plant growth substances on the induction of adventitious buds and the in vitro regeneration. The results showed that the optimal medium for the induction of adventitious buds from Lactuca sativa L. var. ramosa Hort.,Lactuca sativa L. var. crisha Hort. and Lactuca sativa var capitata L. were MS+0.1 mg/L 6-BA+0.2 mg/L NAA,MS+0.1 mg/L 6-BA+0.1 mg/L NAA,and MS+0.2 mg/L 6-BA+0.1 mg/L NAA,respectively. The optimum rooting medium for the three kinds of Lactuca sativa were 1/2 MS+0.05 mg/L NAA,1/2 MS,and 1/2 MS+0.1 mg/L NAA. By analyzing the factors that affect the establishment of high-frequency regeneration system of three kinds of Lactuca sativa,it laid a foundation for further development of functional vegetables by using genetic engineering technology.
