冰菜铝激活苹果酸转运蛋白基因家族的全基因组鉴定与表达分析

Genome-wide identification and expression analysis of the aluminum-activated malate transporter gene family in ice plant

摘要: 为阐明铝激活苹果酸转运蛋白(ALMT)基因家族在盐生植物冰菜(Mesembryanthemum crystallinum L.)中的系统进化与功能特征,在全基因组范围内对冰菜ALMT基因家族成员进行系统鉴定,并分析其在盐胁迫下的表达模式。结果表明,冰菜基因组中共鉴定出9个McALMT基因,不均匀分布于5条染色体上。McALMT蛋白等电点差异较大(6.54~9.20),均不含信号肽,含有5~18个跨膜结构域,主要定位于质膜。系统进化分析表明,McALMT与拟南芥ALMT(AtALMT)蛋白共同聚为5个分支,同一分支内的成员具有相似的基因结构和保守基序。启动子区富含盐胁迫和植物激素响应等顺式作用元件。表达谱分析显示,McALMT2在根、茎、叶中均有表达,且在茎和叶中表达量较高;McALMT1McALMT8McALMT9表现出明显的茎组织偏好性。在盐胁迫下,McALMT4在所有时间点均维持较高表达水平,而McALMT1McALMT2McALMT7在盐胁迫5 d时表现出明显的时间动态响应模式。

Abstract: To elucidate the phylogenetic and functional characteristics of the aluminum-activated malate transporter (ALMT) gene family in the halophyte ice plant (Mesembryanthemum crystallinum L.), a genome-wide systematic identification of ALMT gene family members in ice plant was conducted, and their expression patterns under salt stress were analyzed. The results showed that a total of 9 McALMT genes were identified in the ice plant genome, unevenly distributed on 5 chromosomes. The isoelectric points of McALMT proteins varied greatly (6.54 to 9.20), all lacked signal peptides, contained 5 to 18 transmembrane domains, and were mainly localized to the plasma membrane. Phylogenetic analysis showed that McALMT and Arabidopsis ALMT (AtALMT) proteins were clustered into 5 clades, and members within the same clade had similar gene structures and conserved motifs. The promoter regions were rich in cis-acting elements such as those responding to salt stress and plant hormones. Expression profile analysis showed that McALMT2 was expressed in roots, stems, and leaves, with higher expression levels in stems and leaves; McALMT1, McALMT8, and McALMT9 exhibited obvious stem tissue preference. Under salt stress, McALMT4 maintained a relatively high expression level at all time points, while McALMT1, McALMT2, and McALMT7 exhibited an obvious temporal dynamic response pattern at 5 days of salt stress.
