外源褪黑素对铜胁迫下小麦萌发和幼苗生理特性的影响

Effects of exogenous melatonin on seed germination and seedling physiology characteristics of wheat under copper stress

摘要: 为明确褪黑素在小麦(Triticum aestivum L.)萌发期对铜(Cu)胁迫耐受性的调节作用,以商麦1619和小偃15为试验材料,测定不同浓度(0、50、100、1 000 μmol/L)褪黑素引发处理后在不同Cu2+胁迫浓度(100、150 μmol/L)下种子萌发指标和幼苗生理指标的变化。结果表明,随着外源褪黑素的增加,2个供试小麦品种的发芽势、发芽率、芽长、根长、幼苗超氧化物歧化酶(SOD)活性和过氧化物酶(POD)活性呈先增后减的变化趋势,幼苗丙二醛(MDA)含量呈先减后增的变化趋势。隶属函数分析表明,100 μmol/L Cu2+胁迫下,100 μmol/L褪黑素引发下综合评价值最大,其次为50 μmol/L褪黑素;150 μmol/L Cu2+胁迫下,50 μmol/L褪黑素引发下综合评价值最大,其次为100 μmol/L褪黑素。综上可知,50、100 μmol/L褪黑素对小麦受100、150 μmol/L的Cu2+胁迫有较好的缓解效果。

Abstract: In order to clarify the regulatory effect of melatonin on the tolerance of wheat (Triticum aestivum L.) under copper (Cu) stress at germination stage, Shangmai 1619 and Xiaoyan 15 were used as experimental materials to determine the changes of seed germination indexes and seedling physiological indexes under different Cu stress intensities (100 and 150 μmol/L) after different concentrations (0, 50, 100 and 1 000 μmol/L) of melatonin priming. The results showed that with the increase of exogenous melatonin, the germination potential, germination rate, bud length, root length, and the activities of superoxide dismutase (SOD) and peroxidase (POD) in the seedings of the two wheat varieties increased first and then decreased, and the malonaldehyde (MDA) content of the seedlings decreased first and then increased. The membership function analysis showed that, under 100 μmol/L Cu2+ stress, the comprehensive evaluation value of 100 μmol/L melatonin treatment was the highest, followed by 50 μmol/L melatonin; under 150 μmol/L Cu2+ stress, the comprehensive evaluation value of 50 μmol/L melatonin treatment was the highest, followed by 100 μmol/L melatonin. In summary, 50 and 100 μmol/L melatonin had better mitigation effects on relieving Cu stress under 100 and 150 μmol/L in wheat.
