冬凌草总黄酮超高压辅助低共熔溶剂提取及抗氧化特性

Study on super high pressure assisted deep eutectic solvent extraction of total flavonoids from Rabdosia rubescens (Hemsl.) Hara and its antioxidant activity

摘要: 为优化冬凌草(Rabdosia rubescens (Hemsl.) Hara)总黄酮超高压辅助低共熔溶剂(Deep eutectic solvent,DES)提取工艺,并对其抗氧化活性进行分析。通过单因素试验和响应面试验优化提取工艺条件,对提取的黄酮对1,1-二苯基-2-苦肼基(DPPH)和 2,2′-联氨-双(3-乙基苯并噻唑啉-6-磺酸)二铵盐(ABTS)自由基的清除活性进行分析。结果表明,超高压辅助DES提取总黄酮的最佳条件为料液比1∶40(g/mL)、DES含水量30%、提取压力402 MPa和提取时间5 min,该条件下冬凌草总黄酮得率为9.21%。制备得到的冬凌草总黄酮对DPPH·、ABTS·清除作用的半抑制浓度分别为0.136 mg/mL和0.123 mg/mL。研究可为冬凌草总黄酮开发及抗氧化活性研究提供理论支撑。

Abstract: To optimize the extraction process of total flavonoids from Rabdosia rubescens (Hemsl.) Hara in super high pressure assisted deep eutectic solvent (DES) and analyze its antioxidant activity, the extraction condition was optimized by single factor experiments and response surface experiments. The scavenging activity of the extracted flavonoids against 1,1-dipyl-2-picrylhydrazyl (DPPH) and 2,2′-azinobis (3-ethylbenzothiazoline-6-ulfonic acid) diammonium salt (ABTS) free radicals was analyzed. The results showed that the optimal condition for the extraction of total flavonoids in super high pressure assisted deep eutectic solvent was material to liquid ratio of 1∶; 40 (g/mL), water content in DES of 30% ,extraction pressure of 402 MPa, and extraction time of 5 min, under which the yield of total flavonoids from the Rabdosia rubescens (Hemsl.) Hara was 9.21%. The half-inhibitory concentrations of the prepared total flavonoids of Rabdosia rubescens (Hemsl.) Hara for the scavenging effects of DPPH· and ABTS·were 0.136 mg/mL and 0.123 mg/mL, respectively. The research could provide theoretical support for the development of total flavonoids of Rabdosia rubescens (Hemsl.) Hara and the study of antioxidant activity.;
