谷子EIN3/EIL基因家族鉴定及其在多组织中的表达分析

Identification of the EIN3/EIL gene family in foxtail millet and its expression analysis in multiple tissues

摘要: 为了系统解析谷子(Setaria italica)EIN3/EIL(SiEIN3/EIL)基因家族的进化特征、结构特点及其在生长发育与环境适应中的潜在调控功能,基于谷子全基因组数据对其进行系统鉴定与多组织表达谱分析。通过HMMER与BLASTP联合检索,共鉴定出4个谷子EIN3/EIL家族成员,基因ID分别为Seita.1G205600.1、Seita.7G121700.1、Seita.2G437500.1、Seita.9G419900.1。生物信息学分析表明,该家族蛋白均定位于细胞核且呈亲水性,分散分布于4条染色体上;所有成员均含有外显子,共享6个核心保守基序,且启动子区域富含光响应、多种植物激素响应(如茉莉酸甲酯、脱落酸等)及非生物胁迫响应等顺式作用元件。多组织转录组数据表达分析显示,各基因的表达水平存在差异,其中,Seita.9G419900.1在拔节期至开花期基因表达量明显高于苗期等营养生长阶段,其主要在生殖发育及花器官形成中发挥核心调控作用。

Abstract: In order to systematically elucidate the evolutionary characteristics, structural features, and potential regulatory functions of the EIN3/EIL (SiEIN3/EIL) gene family in foxtail millet (Setaria italica) during growth, development, and environmental adaptation, a systematic identification and multi-tissue expression profile analysis were conducted based on the whole-genome data of foxtail millet. Through combined HMMER and BLASTP searches, a total of four EIN3/EIL family members were identified in foxtail millet, with gene IDs of Seita.1G205600.1, Seita.7G121700.1, Seita.2G437500.1, and Seita.9G419900.1. Bioinformatics analysis indicated that all proteins of this family were localized in the nucleus and were hydrophilic, distributed dispersedly across four chromosomes; all members contained exons, shared six core conserved motifs, and their promoter regions were enriched with cis-acting elements associated with light responses, multiple phytohormone responses (e.g., methyl jasmonate, abscisic acid), and abiotic stress responses. Expression analysis of multi-tissue transcriptome data revealed differences in the expression levels among the genes, among which Seita.9G419900.1 exhibited significantly higher expression from the jointing stage to the flowering stage than during vegetative growth stages such as the seedling stage, playing a core regulatory role mainly in reproductive development and floral organ formation.
