Optimization of enrichment and purification process of puerarin in the production of pueraria starch

Abstract: The method of puerarin enrichment and purification from water discharged by producing pueraria starch using macroporous resin as adsorption material was studied. The saturated capacity of dynamic adsorption (SCD) and elution rate (ER) of fifteen type of resin for total isoflavones of pueraria was investigated. The results indicated that the SCD for total isoflavones of pueraria was 39.41 mg/mL resin and the ER of resin was 91.42% eluted by 70% ethanol aqueous at 1.0 BV/h flow rate. And 4.7 g of crude isoflavones with the content of total isoflavones of 61.67% (UV spectrophotometry) and the content of puerarin of 31.80% (HPLC) could be recovered from 3 L discharge water. A high purity sample which contained 96.37% puerarin was obtained by 5% HCl hydrolysis and recrystallization, and recovery of puerarin was 15.91%. This was the best process optimization for puerarin enrichment and purification from water extracted pueraria starch.
