Identification of two milk proteins of sow by mass spectrometry

Abstract: In order to identify several protein components of porcine milk, whole milk samples of large white pigs (n=8) were collected, and skim milk was obtained by centrifugation. SDS-PAGE was employed to separate skim milk proteins, and tree protein bands on SDS-gel were cut off, followed by treatments of reductive alkylation and trypsin digestion. The resulting peptides were loaded on LC-MS/MS to identify the three proteins with low molecular weight. Two proteins including β-lactoglobulin (β-LG) and whey acidic protein (WAP) were identified successfully. WAP may probably affect porcine milk functions.
