Separation and identification of endophytic bacteria from Polygonatum sibiricum

Abstract: Using Polygonatum sibiricum collected from Ziwuling Nature Reserve in Longdong as material, an endophytic bacterium H-3 with antibacterial activity and genetic stability against Staphylococcus aureus was isolated from its tuber by tissue block separation and agar drilling method. The strain was identified as Bacillus circulans by genomic DNA extraction, 16S rDNA gene cloning and sequencing analysis and morphological characteristics. The determination of solid polysaccharide extracted from fermentation broth showed that the polysaccharide content reached 24.00%.
