Study on high efficient propagation system of Rosa chinensis ‘Red and Pink’

Abstract: This study describes an effective regeneration system via calli induction, somatic embryogenesis and embryos germination using leaflets and compound leaf as explants in Rosa chinensis ‘Red and Pink’. The results showed that, high-frequency primary somatic embryos inductions were obtained from petioles of compound leaf combining 2, 4-D, that L-p and as AgNO3 promoted somatic embryogenesis. When explants were incubated on MS medium supplemented with various concentration of 2, 4-D (3.0, 4.0, 5.0, 6.0 mg/L) respectively, added 400 mg/L L-p and 10 mg/L AgNO3 and solidified using 0.3% Gel, after 16 weeks culture, they firstly formed embryogenic calli and finally somatic embryos. MS medium containing 1.0 mg/L 2, 4-D, 0.05 mg/L 6- BA and 3% glucose was found to be the most effective in the proliferation of somatic embryos. After germination culture on medium(1/2 MS+3% Glucose+0.3% GEL+1.0 mg/L BA+0.01 mg/L NAA), a somatic embryogenesis based regeneration protocol were developed and it will be significant to accelerate the propagation and extensive utilization of Rosa genus species.
