    Evaluation of the correlation between the content of polysaccharides and heavy metals in Polygouatum cyrtouema Hua. and soil in different years

    In order to determine the polysaccharides and heavy metals content of Polygouatum cyrtouema Hua. raw and processed products from two large-scale Huangjing planting bases in Chongyang County, Xianning City, Hubei Province in different years, as well as the corresponding soil heavy metals content in the planting environment, 10 Polygouatum cyrtouema Hua. raw products and 10 soil samples from different years were collected,according to the 2020 edition of the Pharmacopoeia of the People’s Republic of China, the raw medicine Polygouatum cyrtouema Hua. was processed and the polysaccharides content in the processed product was determined using the sulfuric acid anthrone method. The content of heavy metal elements in Polygouatum cyrtouema Hua. and corresponding soil was determined using inductively coupled plasma mass spectrometry. The results showed that as the growth years of Polygouatum cyrtouema Hua. increased, the polysaccharides content of Polygouatum cyrtouema Hua. showed a trend of first increasing and then decreasing. When comparing the polysaccharides content of different years, it was found that the polysaccharides content of Polygouatum cyrtouema Hua. aged 3 or 4 years was higher;after processing, the heavy metals content of Polygouatum cyrtouema Hua. decreased to varying degrees;there was a difference in the heavy metals content in the soil of these two planting bases. Some planting bases had slight pollution of cadmium elements in the soil, while other heavy metal elements were within the safe range. Taking into account factors such as the growth cycle, heavy metals content, and polysaccharides content of Polygouatum cyrtouema Hua., the optimal harvesting period for Polygouatum cyrtouema Hua. was the third or fourth year, and the quality of Polygouatum cyrtouema Hua. was relatively good.

    Detection and drug resistance analysis of virulence and adhesion factor genes of Escherichia coli from Tibetan piglets with diarrhea under house feeding conditions

    To effectively prevent and treat diarrhea in Tibetan piglets, PCR and Kirby Bauer paper diffusion methods were used,the virulence genes, adhesion factor genes and drug resistance of 102 strains of Escherichia coli from Tibetan piglets with diarrhea in Linzhi, Xizang were detected. The detection results of virulence genes and pili adhesion factor genes in Escherichia coli showed that the pili adhesion factor gene with the highest detection rate was F18 (45.10%), followed by F4 (15.69%), and no F41 gene was detected in all strains; among the 102 strains, the pili adhesion factor gene was not detected in 37 strains. The gene with the highest detection rate among virulence genes was Stx2e (25.49%), followed by the STbEAST-1 gene (21.57%) and the STaSTb gene (17.65%). The detection results of Escherichia coli pili adhesion factor genes and non pili adhesion factor genes showed that 44.73% of the strains containing AIDA-1 gene had pili adhesion factor genes (F18, F4), and 54.17% of the strains containing paa gene had pili adhesion factor genes (F18, F4);no F4 gene was detected in the strains that the eae gene was detected. Escherichia coli exhibited high drug resistance to chloramphenicol, tetracycline, ampicillin, and streptomycin, with 86.27% of the total bacterial strains exhibiting resistance to three or more antibiotics.

    Research on ecological polyculture model of seepweed-black sea bream-philippine clam

    In order to explore a seawater ecological polyculture model, three culture objects of Seepweed (Suaeda glauca Bunge), black sea bream (Sparus macrophslus) and Philippine clam(Ruditapes philippinarum)were selected. By setting up seepweed-philippine clam, seepweed-black sea bream, seepweed-black sea bream-philippine clam and black sea bream-Philippine clam four types of polyculture models (black sea bream fry breeding density is 155 fish/m3, Philippine sea clam culture density is 75 ind./m2) with the water quality ind.icators and cultivation of each system monitoring during the breeding process, we try to explore the characteristics of each model. The results showed that the seepweed-black sea bream system and the Seepweed-Philippine clam system have strong water self-purification ability. The ammonia nitrogen, phosphate, and nitrite nitrogen contents of the water body all show a downward trend in the later stage of the experiment, and the two systems show better results. However, the water quality ind.ex of the two systems of Philippine clam and black sea bream polyculture was poor, the mortality of the breeding subjects was high and the system lacks stability. The black sea bream of 155 fish/m3 and Philippine curtain clams of 75 ind./m2 density were suitable for the establishment of Seepweed-black sea bream or Seepweed-Philippine clam polyculture model. Seepweed in the system can not only play a role in purifying water, but also increase the dissolved oxygen content of aquaculture water, and stabilize the pH of aquaculture water.

    The acquisition methods of selenized Codonopsis pilosula polysaccharides were summarized, including the biological conversion method and the chemical synthesis method. The biological conversion method was more suitable for the agricultural production of Codonopsis pilosula medicinal materials. In the chemical synthesis method, the nitric acid-sodium selenite approach assisted by microwave technology significantly improved selenization efficiency and was more suitable for industrial production. This review covered the structural characteristics of selenized Codonopsis pilosula polysaccharides,such as chemical composition, physicochemical properties, and structural characterization,as well as advances in the study of their biological activities, including antioxidant, antitumor, and immunomodulatory effects. It was suggested that future research should focus on optimizing preparation processes, establishing quality control standards and evaluation systems, broadening the scope of biological activity studies, and enhancing product development efforts.

    A g.101284 C>T mutation site was found in intron 9 of the NARS2 gene in the selenium black pig population. After genotyping using PCR product direct sequencing, association analysis was conducted with 23 phenotypes of carcass and meat quality traits.The results showed that the g.101284 C>T locus of the NASR2 gene significantly affected the skin thickness of pig carcasses (P<0.01), and significantly affected the eye muscle area, drip loss for 48 hours, and intramuscular fat content traits of pork (P<0.05). The skin thickness of individuals with CC genotype was significantly lower than that of individuals with TT genotype (P<0.01);the 48 hours drip loss and intramuscular fat content of individuals with CC genotype were significantly higher than those with other genotypes, while the carcass eye muscle area of individuals with TT genotype was significantly smaller than that of individuals with other genotypes (P<0.05). Among these four personality traits, there was a certain degree of correlation between other traits except for skin thickness and eye muscle area traits, which were not significantly correlated. Among them, the 48 hours drip loss had the highest correlation with intramuscular fat content traits (r2=0.882, P<0.01). Therefore, this marker should be reasonably utilized in breeding applications in conjunction with breeding objectives, and individuals of different genotypes should be selected or eliminated for different main selected traits.

    In order to study the differences in volatile flavor compounds and fatty acids composition of beef from different parts of Guanling cattle in Guizhou Province, a comparative analysis was conducted on the volatile flavor compounds and fatty acids composition in six parts of Guanling cattle eye meat, tenderloin, hip meat, beef brisket, rib, and upper brain. The results showed that a total of 68 main volatile compounds were identified in beef from different parts, with terpenes, and alkanes accounting for over 75.000%, which were important factors affecting beef flavor. Fatty acids were important precursors of volatile components, and their content directly affected the taste and flavor of beef. A total of 21 types of fatty acids were detected in beef, including 10 saturated fatty acids (SFA), 4 monounsaturated fatty acids (MUFA), and 7 polyunsaturated fatty acids (PUFA);the content of fatty acids varied in different parts of the body, with unsaturated fatty acids accounting for 43.017%, 39.021%, 48.869%, 46.730%, 40.678%, and 45.147% of fatty acids in eye meat, tenderloin, buttocks, beef brisket, rib ribs, and upper brain, respectively.

    Taking Luoyang city, Henan Province as the research area, four indicators including water conservation, soil and water conservation, biodiversity, and windbreak and sand fixation were selected to evaluate the importance of ecosystem services. Three indicators of soil erosion, land desertification, and rocky desertification were selected for ecological sensitivity evaluation. Using GIS technology, the spatial distribution of each single factor was firstly analyzed, and then the spatial distribution of ecosystem service importance and ecological sensitivity was obtained by superposition and synthesis. Finally on the basis of regional dominant function combined with the current situation of ecological zoning, the region was divided into 6 areas, including water conservation, soil and water conservation and biodiversity maintenance important area, soil erosion prevention and control area, rocky desertification control area, land desertification control area, urban ecological control area and ecological restoration area. The corresponding ecological protection suggestions were put forward according to the different characteristics of each region.

    The chemical components of Angelica sinensis Radix- Cinnamomi cortex was identified, and the anti-inflammatory mechanism of the herb pair was predicted in combination with network pharmacology. GC-MS was used to analyze the volatile components of the herb pair, and then the potential targets of the active components were predicted through the TCMSP database and the Swiss Target Prediction database; inflammation-related gene targets were got through TTD database and GeneCards database; Cytoscape 3.7.2 software was used to construct a drugs-components-targets network diagram; string database and Cytoscape 3.7.2 software were adopted to build a target PPI network, then screening out key targets; the Clsuter Profiler program package in R 3.6.1 software was used to perform gene ontology (GO) function enrichment and KEGG pathway enrichment analysis for potential targets. The results showed that 35 kinds of volatile components were identified from the herb pair, accounting for 70.53% of the total volatile components. From the screening, 33 active compounds, 35 key targets, 1 296 biological processes, and 155 signal pathways were obtained. According to the prediction of network pharmacology results, the herb pair may regulate lipids and atherosclerosis, human cytomegalovirus infection, IL-17 and other signaling pathways through active ingredients such as β-caryophyllene, α-humulene, and α-pinene, and key targets such as TNF-, IL-6, and IL-1B, thereby synergistically exerting anti-inflammatory effects.

    The digital transformation of rural governance was an inevitable choice in line with the development of rural revitalization in the new era. The internal driving force behind this transformation stemmed from the interactive logic among national strategic orientation, the penetration of digital technology, and the decline of traditional governance models. However, the transformation process faced three practical challenges: unequal distribution of digital resources leading to imbalance in governance participation, tension between the rationality of technological tools and local needs, and data security vulnerabilities and the sustainability of governance. To further advance the modernization of China’s rural governance system and governance capabilities, it was necessary to explore the digital transformation path of rural governance from the perspectives of technology, culture, and organization, based on the logic of the mutual construction of technology and society, in order to enhance the efficiency of rural governance and provide support for the systematic implementation of the rural revitalization strategy.

    Effects of short-term flue-cured tobacco-maize rotation on soil enzyme activities, microbial community structure, and substance metabolism

    Mountainous yellow-brown soil in the Wuling Mountain area was studied with four treatments: continuous flue-cured tobacco cropping(KKK), flue-cured tobacco-maize-flue-cured tobacco rotation(KYK), maize-flue-cured tobacco-maize rotation (YKY), and continuous maize cropping(YYY), to investigate the effects of short-term flue-cured tobacco-maize rotation on soil chemical properties, enzyme activities, microbial community structure, and substance metabolism. The results showed that rotation treatments had no significant effects on soil alkali-hydrolyzable nitrogen, available phosphorus, available potassium, or organic matter content, but significantly altered soil enzyme activities. The KYK treatment significantly increased cellulase activity, whereas the YKY treatment significantly decreased urease and sucrase activities. Analysis of the microbial community structure indicated that rotation altered the community structure and diversity of bacteria and fungi. The KYK treatment increased the relative abundance of Acidobacteria and Actinobacteria, while the YKY treatment elevated the abundance of Bacteroidota and Proteobacteria. In fungal communities, rotation treatments increased the relative abundance of Ascomycota and Mortierellomycota. Metabolomic analysis revealed that the KYK treatment significantly increased the contents of diterpenoids and fatty acid amides. Correlation analysis demonstrated a positive relationship between sucrase activity and bacterial diversity, and carbohydrate metabolites were closely associated with the microbial community structure. Short-term flue-cured tobacco-maize rotation could alleviate continuous cropping obstacles by regulating soil enzyme activities, microbial community structure, and substance metabolism, thereby promoting the coordinated and sustainable development of tobacco and grain production in the Wuling Mountain area.

    Using Bacillus amyloliquefaciens TF28 as the host, and by integrating genomic and transcriptomic data, 12 upstream endogenous promoters from highly expressed genes were screened. By constructing a GFP reporter system and evaluating the activity of each promoter in Bacillus, it was found that the promoter strengths of PspoVG, PqoxC, PqoxB, and Pgpd were higher than that of the commonly used promoter P43. When driven by the PspoVG promoter, the transcriptional level of the fenC gene was 4.62 times that of the P43 promoter, and the fengycin yield increased from 84.3 mg/L to 101.2 mg/L. Regarding promoter structure, the sequences of the -35 and -10 regions of PspoVG were highly consistent with the consensus sequences of typical strong promoters, which might be the structural basis for its high-efficiency transcriptional drive.

    Based on the three-dimensional ecological footprint model, this study selected the footprint breadth and ecological footprint depth of nine provinces and two cities in the Yangtze river economic belt from 2007 to 2017 as the research focus, and analyzed the dynamic characteristics of natural capital in the Yangtze river basin by using indicators such as utilization rate of capital flow and utilization ratio of stock flow. The results show that the three-dimensional ecological footprint of the Yangtze river economic belt is increasing, the change rate of footprint breadth and footprint depth is generally changing in the same direction, and the fluctuation gradually tends to be stable after 2015, and the footprint breadth and footprint depth continue to increase at a stable speed after rapid improvement, that is to say, the development level of the Yangtze river economic belt has reached a new level since 2013, and the stock and flow of natural resources demand is higher and increasing. The consumption patterns of flow capital and stock capital are different in different geographical environment regions. The energy consumption among regions is unbalanced and the land use among different regions is unreasonable. Cultivated land and grassland are facing severe situation.

    Based on the agricultural statistical data from 1990 to 2019, the temporal and spatial distribution and “non grain-oriented” situation of crop planting structure in Jiangsu Province in the past 30 years was discussed by using the methods of spatial data analysis and mathematical statistical analysis. The results showed that the planting area of crops in Jiangsu Province had been declining in recent 30 years, and the proportion of the total sown area of cash crops in the total area of crops in Jiangsu Province showed a rapid reduction trend. The sown area of grain crops in northern Jiangsu Province maintained a gradual growth trend, and the“non grain-oriented” trend in southern Jiangsu Province was obvious. Wheat had gradually replaced rice as the food crop with the widest sowing area in Jiangsu Province. Rice planting in southern Jiangsu Province has shifted to central and northern Jiangsu Province in recent 10 years. Corn was the most widely planted in Nantong City, and the sowing level of soybean in the whole province continued to be low. The planting types in Jiangsu were gradually diversified, and a combination of many kinds of crops had been formed. The food security situation had changed for the better.

    Microplastics were plastic particles with a size of smaller than 5 millimetres, which were abundant and persistent in soil, posing a serious global environmental problem. Microplastics entered the crop body through the roots or leaves and affected different stages of crop growth. The pathways of microplastics entering the crop body and the detection methods of microplastics in crops of existing research were summarized, and the effects of different microplastics on crop germination rate, biomass, photosynthesis, oxidative stress system, nutrients, gene expression, etc. were explored. It was found that the influence of microplastics on crops varied with the characteristics of microplastics, crop species and environmental factors. Under certain experimental conditions, microplastics had no effect or a positive effect on crops. However, more studies had shown that microplastics could inhibit crop growth and ultimately affect crop yield. Moreover, microplastics could be enriched upward through food chains and affect human health.

    Chemical composition and antimicrobial activity of the essential oil from Rhododendron capitatum leaves

    The essential oil was extracted by steam distillation, and the components of essential oil were qualitatively analyzed by GC-MS combined with GC retention index. The bacteriostatic effect of Rhododendron essential oil on 9 kinds of pathogenic bacteria was preliminarily studied by filter paper diffusion method. The results showed that 82 compounds were identified in January, 58 in June and 25 in October. The essential oil of leaf of Rhododendron capitatum has certain inhibitory effects on Micrococcus lutea, Staphylococcus aureus, Streptococcus hemolytis-β, Bacillus subtilis and Salmonella paratyphy B. The essential oil components in the leaves of Rhododendron capitatumin different seasons are quite different, which is a reference for the development and utilization of the essential oil in the leaves of Rhododendron capitatum.

    Based on DPSIR model, the driving force - pressure - state - impact - response evaluation index system was established, and TOPSIS evaluation method was used to evaluate the land ecological security in Xinjiang from 2008 to 2019. The results showed that from 2008 to 2019, the overall level of land ecological security in Xinjiang presented a slow upward trend, the level of land ecological security was gradually improved, and the land security status was upgraded from an unsafe level to a relatively safe level. From the perspective of subsystems, the driving force system of Xinjiang land ecological security changed from unsafe state to relatively safe state, the state system, influence system and response system changed from unsafe state to safe state, and the pressure system showed a downward trend from a safe state to an unsafe state. From 2008 to 2019, the overall level of land ecological security in all prefectures, cities and districts in Xinjiang showed a slow upward trend, and the level of land ecological security gradually improved. There were obvious regional differences in the land ecological security situation, and the land ecological security situation in northern Xinjiang was higher than that in southern Xinjiang on the whole.

    Using the leaf,stem,bud, and root of Aquilaria sinensis(Lour.) Gilg. as experimental materials, an orthogonal experimental design was employed via DPS software. By applying different types and concentrations of plant growth regulators (6-BA, 2,4-D, TDZ, NAA, KT), the effects of growth regulators combinations and their concentrations on callus induction, differentiation, and rooting of Aquilaria sinensis were investigated. The results showed that the optimal explant for inducing Aquilaria sinensiscalluswas theleaf, with the highest induction rate of 100%. The optimal medium for leafcallus induction was WPM + 6-BA 1.5 mg/L + 2,4-D 0.5 mg/L + NAA 0.8 mg/L + KT 0.2 mg/L + TDZ 1.0 mg/L. The optimal medium for adventitious bud induction ofAquilaria sinensiswas WPM + 6-BA 0.5 mg/L + NAA 0.2 mg/L + sucrose 20 g/L + agar 6 g/L. The callus from buds and stems could differentiate into adventitious buds. The optimal medium for adventitious bud induction was WPM + 6-BA 0.5 mg/L + NAA 0.2 mg/L + sucrose 20 g/L + agar 6 g/L, resulting in a bud induction rate of 72.22% and an average of 5.68 adventitious buds per explant. The optimal medium for root induction was 1/2 MS + NAA 1.0 mg/L + sucrose 20 g/L + agar 6 g/L, achieving a rooting rate of 46.67%.

    Dendrobium officinal Kimura et Migo has both high edible and medicinal value, which not only contains essential amino acids for human body, but also contains polysaccharides, flavonoids, alkaloids and other bioactive components, these ingredients can reduce blood pressure, blood sugar, anti-tumor and so on. However, it is difficult to identify Dendrobium officinal Kimura et Migo,because of its geographical origin and complex chemical components, use fingerprint technology could provide some theoretical basis to identify and evaluate Dendrobium officinal Kimura et Migo. In addition, domestic and foreign scholars had been isolated and identified polysaccharides, flavonoids, alkaloids and other bioactive components, the effect of these bioactive components was confirmed and their physiological mechanism was deduced. The relevant research on bioactive components and its function mechanism of Dendrobium officinal Kimura et Migo in recent years were summarized,in order to provide some scientific basis for identifying and drug developing of Dendrobium officinal Kimura et Migo.

    Traditional villages carry people's memory and homesickness, as well as hundreds of thousands of historical stories. Studying the narrative of village space is for better memory. Taking Zhangdian village, Jia county, Henan province as an example,the main narrative elements of its place space was explored, the path of its events and space was analyzed, the space syntax theory and depth map software Depthmap were used to quantitative analysis the narrative space, and some corresponding suggestions for the protection of the traditional villages were puts forward.

    The functional degradation and governance countermeasures of the hollow village community

    On the basis of previous studies, the degradation of production function, living function, service function and organization coordination function of the hollow village community was analyzed. And then the driving forces of functional degradation in the hollow village community from three aspects of government power, market power and rural residents’ power were discussed. The function degradation mechanism of the hollow village community was analyzed by push-pull theory, collaborative model and cycle accumulation principle. Finally, some governance countermeasures of functional degradation in the hollow village community were put forward, including respecting farmers’ participation rights and subjectivity, improving rural endogenous development ability, strengthening rural public service supply and improving farmers’ organization degree.
