Optimization of extraction process and determination of total flavonoids in leaves of Podocarpus macrophyllus (Thunb.) D. Don from different origin

Abstract: The reflux extraction method was used to extract total flavonoids from the leaves of Podocarpus macrophyllus (Thunb.) D.Don.The effects of extraction method,solvent concentration,extraction volume and extraction time on the content of total flavonoids in Podocarpus macrophyllus (Thunb.) D.Don were investigated by single factor test.The orthogonal test was used to optimize the extraction conditions and the optimum extraction process was selected.The results showed that the optimum extraction process of total flavonoids from the leaves of Podocarpus macrophyllus (Thunb.) D.Don was 75% ethanol,the ratio of material to liquid was 1:30(g:mL),and the reflux extraction time was 0.5 h.The method has good repeatability,stability and feasibility.
